Body language and proxemics.
Usually when two people that know each other talk, they stand closer and seem to be much more relaxed, if such is the case, it is very easy to tell whether two people are close or not. As opposed to what the case is when two strangers have a conversation, there is, usually some distance in between, as if getting closer meant apporaching unknown territory, and also in terms of body language, people are not as relaxed, and can even be stiff, depending on whether or not the situation is uncomfortable.
